The primary antagonist of Thunderbolts* appears to have been fully confirmed in the most recent trailer, which was unveiled at the current CinemaCon. The film, which is scheduled for release in May 2025, centers on a motley crew of former MCU villains who band together after being lured into a lethal mission by Julia Louis-Dreyfus' Valentina. Although many fans are certain that Sentry, played by Lewis Pullman, is the film's main antagonist, the recently made public description of the CinemaCon footage for the project suggests that someone else is taking the lead.

CinemaCon’s Thunderbolts* trailer confirms Val as the real villain
In the previously described Thunderbolts* trailer, the group tries to use a truck to enter a structure.  When they arrive at their location, though, the team—led by Sebastian Stan's Bucky Barnes—discovers that Julia Louis-Dreyfus' Val was the one who forced their meeting to fire them. This reaffirms that she will be the true antagonist of the movie.

According to The hollywood Reporter, the video begins with Bucky Barnes and David Harbour's red Guardian operating a truck, with Wyatt Russell's john Walker, Florence Pugh's Yelena Belova, and Hannah John-Kamen's Ghost positioned in the rear.  Red Guardian discovers that the Winter Soldier plans to smash their car into the targeted structure to create an opening when he asks Bucky about their "secret plan."

The Thunderbolts* are then shown in the video engaging in combat with a flurry of enemy gunmen, only pausing when they hear Valentina's voice over the loudspeakers.  The cunning agent hurls a litany of taunts at the crew as she tells them to come upstairs.

Then Val says, "You are all so cute.  She confirmed her vindictive plan against the squad by saying, "I sent you down there to kill each other, but instead you make nice and form a team."  Val sends an opposing threat, apparently Lewis Pullman's Sentry, which stops an enraged Bucky from launching a swift counterattack.
